Matchmaker Gold Yeast Two-Hybrid System은 Clontech의 새로운 yeast strain (Y2HGold), Aureobasidin A 항생제에 대한 내성을 포함하는 강력한 reporter 구성, 사용하기 편리한 library, 높은 수준의 발현 벡터 등의 조합으로 Matchmaker 제품 라인을 가장 잘 실현한 제품이다.
Background를 강력하게 제거하는 Aureobasidin A
Matchmaker Gold System은 강력하고 치명적인 yeast (S. cerevisiae) 항생제인 Aureobasidin A (AbA)에 대한 내성을 새로운 reporter로 사용하는 유일한 제품이다. Y2HGold yeast 세포에서는 bait-prey 간 진짜 상호작용이 일어나면, AUR1-C 유전자 산물에 의해 세포가 AbA의 존재 하에서 생장할 수 있도록 한다. AbA는 내성이 없는 yeast는 죽기 때문에 background colony는 절대 자랄 수 없다. 따라서 낮은 강도의 일차 screening에서도 아주 정확하고 높은 비율의 진짜 positive clone을 확인할 수 있다. 또한, AbA 내성을 이용한 선별에는 nutritional marker를 단독으로 사용하는 경우 필요로했던 최적화 과정이 필요 없으며, background colony의 간섭없이 screening을 진행할 수 있다.
4 개의 reporter 유전자로 더욱 정확한 상호작용 확인
Protein-protein간 상호작용을 확인하기 위해 HIS3 nutritional reporter 만을 유일하게 사용하는 Yeast two-hybrid system은 수많은 (i) background colony와 (ii) false positive colony를 생성한다. Background colony는 HIS3 발현이 엄격하게 조절되지 않아서 생긴 결과인 반면, false positive은 prey 단백질이 독립적으로 reporter를 활성화시켜 나타나는 현상이다.
Matchmaker Gold의 정확성은 4 개의 selectable reporter gene (AUR1-C, HIS3, ADE2, and MEL1 (α-galactosidase))의 사용 결과이며, 각각의 reporter gene은 3 개의 다른 GAL4-responsive promoters에 의해 유도 발현된다(Figure 1). 엄격하게 조절되는 이러한 reporter의 전략적인 조합을 통하여 사실상 3 개 모두에는 결합하지 못하고 한 개의 프로모터 서열에만 직접 결합한 library prey 단백질에 의해 생성되는 false positive을 효율적으로 제거할 수 있다. Matchmaker Gold는 낮은 background와 높은 정확도로 library를 screening하기 위하여 Aureobasidin A 내성과 4 개의 정확한 reporter를 이용하는 유일한 yeast two-hybrid system이다.
편리한 Media Sets & Colony PCR Mix
Yeast Media Set 2 Plus는 Matchmaker Gold Two-Hybrid 실험에 필요한 pouch 형태의 배지, Aureobasidin A 및 X-α-Gal로 구성된 제품이다. Matchmaker Insert Check PCR Mix 2는 library vector에 포함된 insert cDNA를 yeast colony로부터 직접 증폭할 수 있는 premix 형태의 PCR 효소로 Matchmaker Gold yeast one- & two-hybrid library screening system에 사용하기 위하여 고안되었으며, positive colony로부터 insert cDNA를 빠르게 증폭, 분류 및 분석할 수 있다.
