균일하고 깨끗한 세포 분화가 가능한 기존 DEF-CS 500 culture system (Code Y30010)의 능력을 기반으로, single cell cloning 능력을 향상시킨 제품이다. 본 제품을 사용하면 고효율의 single cell cloning이 가능하며,
DEF-CS 500과 같은 장기간 미분화도 가능하여 균일하게 hiPS cell 배양을 할 수 있다.
Cellartis® iPSC CRISPR/Cas9 Gesicle and Single-Cell Cloning System (Code 632642)는 세포막 유래
nanovesicle의 일종인 Gesicle을 사용하여 Cas9/sgRNA Ribonucleoprotein (RNP)를 세포에 도입하는 CRISPR/Cas9 Gesicle producer system (Code 632613)과 Cellartis® iPSC Single-Cell Cloning DEF-CS™ Culture Media Kit
(Code Y30021)가 포함된 Kit이다 Gesicle을 이용하면 hard to transfect cell line인 human iPS 세포에 대해서도 효율적인 Cas9/sgRNA RNP의
도입이 가능하며, off target effect를 줄일 수 있다

도입에 최적인 Guide-it™ Recombinant Cas9 (Electroporation-Ready) (Code 632641) 및 Cellartis® iPSC Single-Cell Cloning DEF-CS™ Culture Media Kit (Code Y30021)가 포함된 kit이다. 기존의 plasmid 도입에
비해 electroporation을 이용한 Cas9/sgRNA RNP의 도입으로 off-target effect를 줄일 수 있으며, Cas9 단백질의 NLS (Nuclear localization sequence)와
electroporation에 적합한 buffer를 사용하여 고효율의 Cas9/sgRNA RNP 세포 내 도입이 가능하다.
