제품 설명

Lenti-X™ Packaging Single Shots은 4세대 lentivirus packaging system으로, 아주 편리한 one-step 방식으로 실험을 진행하며, 전용 packaging cell을 이용했을 때 일반적으로 10⁷-10⁸ IFU/ml의 고역가 Lentivirus를 생산할 수 있다.

제품은 lentivirus를 packaging 할 수 있는 vector와 transfection 시약이 모두 최적의 비율로 single tube 내에 분주 및 동결건조 되어 있는 형태로, 발현을 원하는 tropism (envelope)을 선택할 수 있다. 또한, single tube 형태와 96 well plate 형태를 선택할 수 있어, high-throughput이 요구되는 실험에도 적용할 수 있다.

VSV-G, ecotropic 외의 tropism을 적용했을 때 더 높은 유전자 도입 효율이 기대되는 경우
Narrowed tropism에서 높은 감염 효율을 보이는 특정한 cell type 적용 시
Envelope 예) Baboon envelope, Cocal envelope, Measles envelope, Mokola envelope, Ebola envelope 등
Target cell 예) 조혈모세포 (hematopoietic cell), 면역세포 (immune cell - T cell, B cell 등), 특정 장기 타겟

Lenti-X Packaging Single Shots (Envelope-Free)는 single shot 내 envelope vector가 포함되어 있지 않고, 별도의 empty vector 형태로 제공된다. 따라서 원하는 envelope protein 서열을 직접 cloning하고, lentiviral expression vector와 함께 single shot에 넣어 virus를 packaging한다. 또한 transduction 효율을 확인할 수 있도록 ZsGreen1와 luciferase를 각각 발현하는 reporter vector 2종이 제품 내 포함되어 있다.

실험 주의사항

- 4세대의 lentivirus packaging system은 Tet-off 기술이 도입되어 있어, packaging cell 배양 시 tetracycline을 포함하지 않는 배지 조건으로 배양해야 한다. Tetracycline-derived contaminants로 인한 background를 최소화할 수 있는 Tet approved FBS의 사용을 적극 권장한다.
- 제품 내에는 packaging cell line을 포함하고 있지 않다. 보다 고역가의 바이러스를 생산할 수 있는 Lenti-X™ 293T Cell Line (Code 632180)를 함께 구매하여 사용할 것을 추천한다.
