Retro-X Concentrator로 retrovirus의 역가를 100 배까지 증가시킬 수 있으며, 더 많은 바이러스를 만들거나, 추가로 packaging 세포에 transfection할 필요 없이 충분히 높은 MOI로
목적세포를 감염시킬 수 있다.

Simple 조작과정: Mix, Wait 그리고 Spin

Retro-X Concentrator는 빠르고, 간편하며 어떠한 종류의 retrovirus stock도 농축시킬 수 있는 아주 효율적인 방법을 제공한다. 우선 retrovirus 상층액을 Retro-X Concentrator 시약과 섞은 후 하룻밤 반응시킨 후 반응액을 일반 원심분리기로 원심분리한다 (Figure 1). 상층액의 양과 viral envelope에 따라 MMLV 또는 MSCV에 기반을 둔 retroivirus의 역가를
100 배까지 증가시킬 수 있으며 (Figure 2), 높은 회수율을 얻을 수 있다. Clontech의 Retro-XSystem의 모든 retroiviral vector 뿐만 아니라 다른 모든 retroivirus의 상층액을
Retro-X Concentrator로 농축할 수 있으며 필요에 따라 용량을 증감할 수 있다.

100 배까지 역가 증가

Retro-X Concentrator을 사용하면 다양한 envelope-pseudotyped retroivirus의 상층액 10 ml을 100 μI로 농축하여 역가를 46~116 배까지 증가시킬 수 있다 (Figure 2).

초고속원심분리보다 더욱 간단한 방법

Retro-X Concentrator과 초고속원심분리 방법을 비교하였을 경우, Retro -X 방법의 장점은 매우 명확하다 (Table 1). Retro-X Concentrator는 초고속원심분리 방법보다 더 쉽고 융통성이 있다. Retro-X Concentrator 시약은 4x 농도의 시약으로 제공되기 때문에 상층액에 포함된 바이러스의 양과 역가에 상관없이 사용할 수 있다. 상층액 250 ml 까지도 적용 가능하다.
