MSCV (Murine Stem Cell Virus) Retroviral Expression System은 murine 또는 인간 조혈세포 (hematopoietic cell), 배아줄기세포(embryonic cell line, ES), 배아암세포 (embryonal carcinoma cell, EC cell)를 포함한 pluripotent cell line에서 목적유전자를 도입하고 발현시키기 위하여 최적화된 vector들로 구성되어 있다. 이들 시스템은 포유류 세포에도 효과적으로 적용될 수 있으며 (1-3), 세포배양과 유전자 도입 분석에 있어서 분화 (development), 배발생 (embryogenesis) 또는 면역반응 (immune response) 과정에 관여하는 유전자의 기능 분석에 이상적이다.

Infection이 어려운 세포를 위한 설계

MSCV System은 pMSCVneo, pMSCVhyg 그리고 pMSCVpuro의 3 가지 vector를 포함하고 있다. 이 vector들은 도입이 어려운 cell line에 적용할 수 있도록 murine stem cell PCMV virus로부터 특별히 제작된 long terminal repeat (LTR)을 가지고 있다. 이 LTR은 ES세포와 EC세포에서 전사활성화를 증진시킴과 동시에 전사억제작용을 방지할 수 있도록 여러 개의 point mutation과 하나의 deletion를 가지고 있다는 점에서 MMLV LTR과는 다르다. 결과적으로, PCMV LTR은 줄기세포나 다른 포유류 cell line에서 목적 유전자가 구조적으로 고발현 되도록 한다. MSCV 시스템은 RetroPack PT67 Packaging Cell Line를 포함하고 있으며, 이 packaging cell lines는 광범위한 포유류 숙주 세포를 충분히 감염시킬 수 있는 고역가의 바이러스를 생산할 수 있다.
